ItemVolunteers in Australian archivesde Villiers, A ; Laurent, N ; Stueven, C (ROUTLEDGE JOURNALS, TAYLOR & FRANCIS LTD, 2017-01-01)Why do volunteers choose to contribute thousands of unpaid hours per week to Australian archives? This paper presents the results of a nationwide web survey that provides insight into the demographics, motivations and experiences of volunteers in Australian archives. The findings provide a representative overview of formal volunteers in Australian archives, determining ‘who’ they are, the value of the contributions they provide and the level of training and support offered to them. This study is a continuation of existing discussions about volunteers in Australian archives and represents an opportunity for the development of stronger relationships with Australian archival volunteers and, through them, the communities our archives serve.

ItemNo Preview AvailableThe major membrane nuclease MnuA degrades neutrophil extracellular traps induced by Mycoplasma bovisMitiku, F ; Hartley, CA ; Sansom, FM ; Coombe, JE ; Mansell, PD ; Beggs, DS ; Browning, GF (ELSEVIER SCIENCE BV, 2018-05-01)Mycoplasma bovis has been increasingly recognised worldwide as an economically important pathogen of cattle, causing a range of diseases, including pneumonia, mastitis, polyarthritis and otitis media. It is believed that M. bovis utilises a range of cell surface proteins, including nucleases, to evade the host immune response and survive. However, despite the importance of neutrophils in controlling pathogenic bacteria, the interaction between these cells and M. bovis is not well-characterised. In addition to phagocytosis, neutrophils combat pathogens through the release of neutrophil extracellular traps (NETs), which are composed of their nuclear and granular components, including DNA. Here we investigated the effect of the major membrane nuclease MnuA of M. bovis, which in vitro is responsible for the majority of the nuclease activity of M. bovis, on NET formation. We quantified NET formation by bovine neutrophils 4 h after stimulation with wild-type M. bovis, an mnuA mutant and a mnuA-pIRR45 complemented mnuA mutant. NETs were detected following stimulation of neutrophils with the mnuA mutant but not after exposure to either the wild-type or the mnuA-pIRR45 complemented mutant, and NETs were degraded in the presence of even low concentrations of wild type M. bovis. Surprisingly, there was no increase in levels of intracellular reactive oxygen species (ROS) production in neutrophils stimulated with M. bovis, even though these neutrophils produced NETs. These results clearly demonstrate that M. bovis can induce NET formation in bovine neutrophils, but that the major membrane nuclease MnuA is able to rapidly degrade NETs, and thus is likely to play a significant role in virulence. In addition, M. bovis appears to induce NETs even though ROS production seems to be suppressed.

ItemThe cationic small molecule GW4869 is cytotoxic to high phosphatidylserine-expressing myeloma cellsVuckovic, S ; Vandyke, K ; Rickards, DA ; McCauley Winter, P ; Brown, SHJ ; Mitchell, TW ; Liu, J ; Lu, J ; Askenase, PW ; Yuriev, E ; Capuano, B ; Ramsland, PA ; Hill, GR ; Zannettino, ACW ; Hutchinson, AT (WILEY, 2017-05-01)We have discovered that a small cationic molecule, GW4869, is cytotoxic to a subset of myeloma cell lines and primary myeloma plasma cells. Biochemical analysis revealed that GW4869 binds to anionic phospholipids such as phosphatidylserine - a lipid normally confined to the intracellular side of the cell membrane. However, interestingly, phosphatidylserine was expressed on the surface of all myeloma cell lines tested (n = 12) and 9/15 primary myeloma samples. Notably, the level of phosphatidylserine expression correlated well with sensitivity to GW4869. Inhibition of cell surface phosphatidylserine exposure with brefeldin A resulted in resistance to GW4869. Finally, GW4869 was shown to delay the growth of phosphatidylserine-high myeloma cells in vivo. To the best of our knowledge, this is the first example of using a small molecule to target phosphatidylserine on malignant cells. This study may provide the rationale for the development of phosphatidylserine-targeting small molecules for the treatment of surface phosphatidylserine-expressing cancers.

METHODS: Two focus groups were conducted with 17 participants to explore informational, psychological, emotional, social, practical and physical needs, 6-30 months post-treatment for lymphoma. Perceptions regarding a potential model of survivorship care were also elicited. RESULTS: Thematic content analysis revealed five key themes: Information; Loss and uncertainty; Family, support and post-treatment experience; Transition, connectivity and normalcy, and Person-centred post-treatment care. Participants described a sense of loss as they transitioned away from regular interaction with the hospital at the end of treatment, but also talked about the need to find a "new normal". Establishing post-treatment support structures that can provide individualised information, support, reassurance and referrals to community and peer support were identified as a helpful way to navigate the transition from patient to post-treatment survivor. CONCLUSIONS: Participants in our study articulated a need for a flexible approach to survivorship care, providing opportunities for individuals to access different types of support at different times post-treatment. Specialist post-treatment nurse care coordinators working across acute and community settings may offer one effective model of post-treatment support for survivors of haematological malignancies.

ItemAntimicrobial stewardship in the dairy industry: responding to the threat of antimicrobial resistance.Coombe, JE ; Tymms, SM ; Humphris, M (Wiley, 2019-07-01)Dairy Australia is the national service body for the Australian dairy industry. Its role is to help farmers adapt to a changing operating environment and achieve a profitable, sustainable dairy industry. Although the use of antibiotics in Australian agriculture is relatively low in global terms, Dairy Australia recognises important drivers for continuous improvement in antimicrobial stewardship (AMS). Dairy Australia's first strategic priority is to support profitable farms. This priority has driven the development of a range of on-farm change management programs in the animal health and welfare fields to optimise the unit cost of production and dairy cattle welfare. Dairy Australia's third strategic priority is to further develop a 'trusted dairy industry'. Previous and current work under these two strategies position the dairy industry favourably with respect to confronting the challenge of antimicrobial resistance (AMR) and supporting sound AMS with the guiding principle of 'as little as possible, as much as necessary'. However, given an incomplete but ongoing threat of AMR, more work is needed. Supported by Dairy Australia, the dairy industry has developed an antimicrobial use strategy aligning with the Australian Animal Sector National AMR Plan 2018.

ItemYou can't be Shakespearean talking about the institutionalisation of sex offenders: Creativity and creative practices of multilingual doctoral writersThurlow, S ; Morton, J ; Choi, J (PERGAMON-ELSEVIER SCIENCE LTD, 2019-03-01)The enigma of creativity is rarely discussed in doctoral education, yet it nestles snugly against the term originality, a key criterion for thesis assessment. This article engages with this occluded topic through an investigation of how four L2/multilingual PhD candidates studying in the Faculty of Arts in an Australian university perceive the presence of creativity in their doctoral writing. It also explores how and when these writers feel they can be creative in their writing practices. Methodological approaches included a workshop program designed around the concept of creativity for Arts doctoral students, followed by individual and group interviews. The findings indicate that while each doctoral writer actively engaged with the idea of creativity they also encountered social, cultural, political and other environmental barriers. These constraints often led to a lack of writer agency which, in turn, led to self-censorship. Nevertheless, several enablers to their creativity were uncovered with participants recognising the usefulness of learning specific writing practices and other strategies to allow creativity to emerge in their work. The article also offers a model of creativity that may provide a useful starting point for others to use in understanding the highly complex role creativity holds for doctoral writing.

ItemConstitutive and Inflammatory Immunopeptidome of Pancreatic beta-CellsDudek, NL ; Tan, CT ; Gorasia, DG ; Croft, NP ; Illing, PT ; Purcell, AW (AMER DIABETES ASSOC, 2012-11-01)Type 1 diabetes is characterized by the autoimmune destruction of pancreatic β-cells. Recognition of major histocompatibility complex (MHC)-bound peptides is critical for both the initiation and progression of disease. In this study, MHC peptide complexes were purified from NIT-1 β-cells, interferon-γ (IFN-γ)-treated NIT-1 cells, splenic and thymic tissue of 12-week-old NOD mice, and peptides identified by mass spectrometry. In addition to global liquid chromatography-tandem mass spectrometry analysis, the targeted approach of multiple-reaction monitoring was used to quantitate the immunodominant K(d)-restricted T-cell epitope islet-specific glucose-6-phosphatase catalytic subunit-related protein (IGRP)₂₀₆₋₂₁₄. We identified >2,000 MHC-bound peptides; 1,100 of these presented by β-cells grown under normal conditions or after exposure to IFN-γ. These include sequences from a number of known autoantigens. Quantitation of IGRP₂₀₆₋₂₁₄ revealed low-level presentation by K(d) (~25 complexes/cell) on NIT-1 cells after IFN-γ treatment compared with the simultaneous presentation of the endogenously processed K(d)-restricted peptide Janus kinase-1₃₅₅₋₃₆₃ (~15,000 copies/cell). We have successfully sequenced peptides from NIT-1 β-cells under basal and inflammatory conditions. We have shown the feasibility of quantitating disease-associated peptides and provide the first direct demonstration of the disparity between presentation of a known autoantigenic epitope and a common endogenously presented peptide.

ItemPhysical and Linkage Maps for Drosophila serrata, a Model Species for Studies of Clinal Adaptation and Sexual SelectionStocker, AJ ; Rusuwa, BB ; Blacket, MJ ; Frentiu, FD ; Sullivan, M ; Foley, BR ; Beatson, S ; Hoffmann, AA ; Chenoweth, SF (GENETICS SOCIETY AMERICA, 2012-02-01)Drosophila serrata is a member of the montium group, which contains more than 98 species and until recently was considered a subgroup within the melanogaster group. This Drosophila species is an emerging model system for evolutionary quantitative genetics and has been used in studies of species borders, clinal variation and sexual selection. Despite the importance of D. serrata as a model for evolutionary research, our poor understanding of its genome remains a significant limitation. Here, we provide a first-generation gene-based linkage map and a physical map for this species. Consistent with previous studies of other drosophilids we observed strong conservation of genes within chromosome arms homologous with D. melanogaster but major differences in within-arm synteny. These resources will be a useful complement to ongoing genome sequencing efforts and QTL mapping studies in this species.
