Abstract
Salmonella infection is a major cause of morbidity and mortality worldwide, and the development of effective vaccines requires a better understanding of the protective immune mechanisms. The importance of CD4+ T cells during Salmonellosis is well established, but antigenic targets, in vivo dynamics and the phenotype of Salmonella-specific CD4+ T cells are less than fully resolved. In this project, four antigenic targets were selected from a novel Salmonella-derived MHC class II immunopeptidome and respective MHC class II tetramers generated, which enabled the direct identification, enumeration and characterization of so far unreported Salmonella-specific CD4+ T cells during murine Salmonellosis. Our results demonstrate the feasibility and utility of using immunopeptidomics and tetramer technology to study Salmonella-specific CD4+ T cell immunity in a murine model of Salmonellosis. Furthermore, the novel antigens and tetramers provide valuable tools for next-generation vaccine development against Salmonella.
